Yes, they will spawn. They are not normal Crimson Guardians, and I didn't stay to try kill them. But I assume they are Master Guardians, the same with those in the Archer Room?

Basically: Don't leave someone AFK in this room.

 

Assuming you're Lv.130+, you'll need 202 accuracy to hit all three bosses on the bottom 100% of the time (Hsalf - the Thief Boss, is the issue).
When it comes to Master Gawyllacs - the Pirate Boss, you need even more than 202 accuracy. The exact number is unknown, as I've not managed to find any data on this guy.

 

This thread never talked much about the bonus stage, so I guess I'll say a few words about it.
The link above takes you to a picture showing all of the chests in the bonus room - numbered. If you're 5 players (which is common) each player will break and loot from the boxes numbered with the same number (example: Bill is given the number 3 by the leader, so he breaks all the boxes numbered "3")

Good practice is : Don't loot Pie if you're a non-mage,
and don't loot Gelt Chocolate if you're a mage. These items often vanish due to poor communication, but it's possible to have some time left over to allow these items to be picked up.

Once one player enters the bonus room, a 1-minute timer will start, so it's important to have coordinated things before this. Before the bonus room is a lobby with a 5 minute timer. This is normally enough time but it's still adviced to coordinate everything in the boss room once the 5 bosses are dead.
